Lipase Production in Solid State Fermentation Using Aspergillus niger: Response Surface Methodology
Authors
Abstract:
Among enzymes, lipases have been widely investigated because of the numerous industrial applications. In this study, optimization of lipase production by Aspergillus niger in solid state fermentation from rice bran as solid substrate was investigated. The optimal conditions with the aid of central composite design (CCD) under response surface methodology (RSM) were obtained. In the analysis of 20 experimental runs, effects of three variables such as olive oil concentration, glucose concentration and humidity ratio on lipase activity were evaluated. The interaction between lipase activity and independent paired parameters were illustrated by three-dimensional diagrams while other factors were set at zero level. The optimum values of oil concentration, glucose concentration and humidity ratio were 1.78, 2.54 and 1.95 % (g/gdss), respectively. Also, the effect of temperature and pH on the relative lipase activity in enzyme reaction was evaluated. The lipase produced was highly active at 30.3°C and pH value of 6.87.

Optimization of Fermentation Medium for Extracellular Lipase Production from Aspergillus niger Using Response Surface Methodology
Lipase produced by Aspergillus niger is widely used in various industries. In this study, extracellular lipase production from an industrial producing strain of A. niger was improved by medium optimization. The secondary carbon source, nitrogen source, and lipid were found to be the three most influential factors for lipase production by single-factor experiments. According to the statistical a...

Central composite orthogonal design was applied to quantify relations of xylanase production, loss of dry matter and change of pH with four critical variables during solid state fermentation of a mixture of wheat bran and wheat straw on which Aspergillus niger CCUG 33991 was cultivated. The studied variables included the percentage of wheat straw, temperature, moisture content, and fermentati...
full textSolid State Fermentation for Production of Chitosan by Aspergillus Niger
The effect of Solid State Fermentation (SSF) on Chitosan production by A. niger was investigated. A. niger BBRC 20004 from Biochemical and Bioenvironmental Research Centre of Sharif University of Technology (Tehran, Iran), was grown on corn residue. Chitosan was extracted from the fungal mycelia using hot alkaline and acid treatment. A. niger was incubated for 12 days on corn residue with moist...

Background and Objectives: Cellulase is an important enzyme with multiple applications in industries, including food, laundry, pharmaceutical, textile, pulp, paper and biofuel industries. Solid-state fermentation (SSF) is a method for cellulase production, which includes several advantages, compared to submerged fermentation. In this study, cellulase was produced by three filamentous fungi, i.e...
full textProduction of Glucoamylase by Aspergillus Niger under Solid State Fermentation
In this study, Glucoamylase production by Aspergillus niger was investigated under solid state fermentation using low cost by-products of agricultural processes as substrate. Highest enzyme production was observed when a combination of wheat bran (WB) and corn flour (CF) was used as compared to WB+ rice bran, WB+ rice flour and WB alone. Several compositions of corn flours were experimented. Th...
full textStudies on the Lipase Production by Aspergillus Niger through Solid State Fermentation
The current study includes isolation and screening of different fungal cultures from soil samples for lipase production by solid state fermentation. Seven different lipolytic fungal strains were isolated and screened for the purpose. These isolates were identified on the basis of morphological and microscopic studies. Among these fungal strains, Aspergillus niger gave best reproducible results ...
